VEGF,即vascular endothelial growth factor,也稱VEGFA或VEGF-A,中文名為血管內(nèi)皮生長(zhǎng)因子,是一種可以強(qiáng)烈誘導(dǎo)血管生長(zhǎng)的因子,在血管過(guò)程中起重要作用。腫瘤細(xì)胞在低氧(hypoxia)情況下可以分泌VEGF。VEGF的7種isoform是VEGF mRNA可變剪接(alternative splicing)的結(jié)果。zui常見(jiàn)的VEGF的isoform包括VEGF 121,VEGF 165,VEGF 189(相當(dāng)于VEGF f、VEGF d和VEGF b)。VEGF的表達(dá)水平和腫瘤內(nèi)的血管生長(zhǎng)情況呈正相關(guān),并且和腫瘤的轉(zhuǎn)移風(fēng)險(xiǎn)呈正相關(guān)。VEGF可以促進(jìn)內(nèi)皮細(xì)胞增殖,促進(jìn)細(xì)胞遷移,并抑制細(xì)胞凋亡。抑制VEGF信號(hào)轉(zhuǎn)導(dǎo)可以抑制多種腫瘤的發(fā)生和發(fā)展。VEGF可以形成同源二聚體,可以和其受體VEGFR1/Flt-1和VEGFR2/Kdr,以及heparan sulfate和肝素相結(jié)合。VEGF-165和VEGF-145可以和Neuropilin-1相結(jié)合。
BACKGROUND
The onset of angiogenesis is believed to be an early event in tumorigenesis and may facilitate tumor progression and metastasis. Several growth factors with angiogenic activity have been described. These include fibroblast growth factors (FGFs), plaet derived growth factor (PDGF) and vascular endothelial growth factor (VEGF). VEGF is a dimeric glycoprotein with structural homology to PDGF. Several variants of VEGF have been described that arise by alternative mRNA splicing. It has been speculated that VEGF may function as a tumor angiogenesis factor in vivo because the expression pattern of VEGF is consistent with a role in embryonic angiogenesis. VEGF mRNA is formed in some primary tumors, VEGF is produced by tumor cell lines in vitro and VEGF mitogenic activity appears to be restricted to endothelial cells. A member of the PDGF receptor family, Flt, has been identified as a high-affinity receptor for VEGF.

APPLICATIONS
VEGF (VG-1) is recommended for detection of the 189, 165 and 121 amino acid splice variants of VEGF of mouse, rat, human and dog origin by Western Blotting (starting dilution 1:200, dilution range 1:100-1:1000), immunoprecipitation [1-2 μg per 100-500 μg of total protein (1 ml of cell lysate)], immunofluorescence (starting dilution 1:50, dilution range 1:50-1:500) and immunohistochemistry (including paraffin-embedded sections) (starting dilution 1:50, dilution range 1:50-1:500).
